Optimization of Fermentation Culture Medium for Grifola frondosa and Extraction of Trehalose

Abstract

The fermentation medium composition of Grifola frondosa was optimized,and the extraction of trehalose from Grifola frondosa was studied.The results showed that the optimum component of medium of Grifola frondosa was:potato 200 g,glucose 40 g,wheat bran 25 g,Chinese chestnut shell 150 g,KH2PO4 3g,MgSO41.5g and water 1 L,under which the optimum weight of dry fungi was 15.1 g/L.The best determination conditions of trehalose content by 0.2% anthrone-sulphuric acid colorimetric method(the proportion of extraction was 1:4)were as follows:absorption peak 625nm and heating time 2 min.For the extraction of trehalose from Grifola frondosa,the optimum extraction times,time,temperature,and 0.5 mol/L Trichloroacetic acid volume were 3,20 min,0℃,and 20 ml,respectively,under which 14.522mg of trehalose was obtained from 1.0 g wet mycelium.
