Abstract

Aim:To study and modify the optimal methods of silver staining of denaturing and non denaturing polyacrylamide gels.Methods:polymerase chain reaction (PCR) polyacrylamide gel electrophoresis silver nitrate (AgNO3) staining were used to detect DNA fragments of both 159~196 bp and 336 bp under conditions of different concentration and temperatnre developed solution.Results:The DNA fragments detected by the modified method of silver staining were clear and suitable to analyze.Conclusion: Using silver staining to analyze the polyacrylamide gel bands contributes to a longterm preservation and can reduce the pollution effects on human beings and environment.
