Abstract
The effects of explants, light conditions as well as different cytokinins (kinetin and BAP) and auxins (2, 4-D, NAA and IAA) on the induction and growth of callus in Linum glaucum were studied. Initially, root, hypocotyl and cotyledon explants were cultured using different cytokinins, either in dark or light for 30 days. Afterwards, the percentage of callus induction, calli diameter and fresh weight were evaluated. Accordingly, the root explants showed better callus induction when cultured on MS medium containing 1 mg/l kinetin. In other experiment, the explants were cultured on MS medium supplemented with 0.5 mg/l of different auxins to determine the most favourable auxin for callus induction and growth. As a result, the highest callus formation was observed on the medium containing 2,4-D, while IAA and NAA were not effective for callus induction and growth. The results of the present study provide a basis for callus induction procedure of this medicinal plant for further investigations.
