Abstract
OBJECTIVE To determine the concentrations of 5-fluorouracil(5-Fu)in human plasma by HPLC.METHODS The detection wavelength was at λ max=254 nm.The mobile phase was H 2O-CH 3OH-CH 3COOH(99.9%)(96.95∶3.0∶0.05),pH=3.5.40 μL of the internal standard 5-Bru(200 μg·mL -1)was added into 200 μL of the plasma sample.Then 400 μL of methanol was added and centrifuged for 5 min.20 μL of the supernatant were injected into the HPLC system.RESULTS The linear range of 5-Fu was 0.66~42.00 μg·mL -1,Y=11.82X+0.078(r=0.999 2) and the recovery was 99.77% with RSD 2.0% (intra-day) and 4.0%(inter-day).CONCLUSION This method appeared bo te simple and accurate.It is suitable for the determing of drug concentrations of 5-Fu in human plasma in clinical use.
