Effect of Benign Prostatic Hyperplasia on the Deposit of Collagen on the Ageing Rat Bladder

Abstract

Objective:To observe the morphologic changement of aged bladder and the effect of benign prostatic hyperplasia(BPH) on the deposit of collagen on the ageing rat bladder.Materials and Methods:Materials:Young(3 months) and aged(18 months) male Wistar rats.In addition to blank youth group and blank elderly group,the other rats were injected testosterone plus surgical castration replicated with BPH model.The model rats were randomly divided into model months old youth,older model group,model group + elderly finasteride group,model elderly group + doxazosin group.The rats were fed for 30 days to give the corresponding drugs.Immunohistochemical staining of bladder tissue with collagen Ⅲ and collagen I.Results:The aged rats significantly decreased detrusor cells,and increased collagen deposition.Detrusor smooth muscle cells in old BPH rats were significantly increased compared with normal detrusor young and old rats,and decreased after treatment by doxazosin and finasteride.Collagen I positive area ratio(18.16±3.89) were higher than that of young group(9.74±1.54,P0.01),lower than older model group(23.30±1.32,P0.01),doxazosin group(23.37±2.94,P0.01),and finasteride group(21.94±3.23,P0.05).collagen Ⅲ positive area ratio of old model group bladder(40.11±2.03) was significantly higher than blank elderly(26.49±3.12,P0.01),doxazosin group(30.88±4.73,P0.01) and finasteride group(26.13±2.46,P0.01).The rate blank elderly group(0.68±0.10) was significantly lower than blank young group(1.30±0.28,P0.01).The rate of collagen Ⅲ/collagen I of Doxazosin group(0.78±0.18,P0.05)and finasteride group(0.82±0.09,P0.01) was significantly higher than the model group(0.58±0.36),but there was no significant differences compared with blank elderly group.Conclusion: The aged change is an important reason for changes in aged BPH rat bladder,simply remove the obstruction cannot improve it.The results suggest that we should give full consideration on the aging changes in the bladder itself in clinical patients with benign prostatic hyperplasia,it is important of early protection of bladder function in order to delay the aging process of change.
