Abstract
Objective:To establish a new method for the determination of astragaloside Ⅳ in radix astragali.Methods:A ELSD-HPLC method was used to determine the content of astragaloside Ⅳ in the test solution.The separation were carried out on a Zorbax SB C18(150mm×4.6mm,3.5μm) column with acetonitrile-water(35∶65) as mobile phase.The flow rate was 1.0mL·min-1 and the tube temperature of the ELSD was set at 110℃.The gas flow was set at 2.5L·min-1.Results:The assay was validated in the weight range of 0.40~1.98μg(r=0.9998).The average recovery was 103.8%(RSD=1.2%).Conclusion:The method is accurate and reproducible,and can be used for determination of astragaloside Ⅳ in radix astragali.
