Abstract

Objective:To establish an HPLC-ELSD method for the determination of the content of astragaloside Ⅰ,astragaloside Ⅱ and astragaloside Ⅳ in Radix Astragali from various habitats.Methods:The conditions of chromatography:ZORBAX Eclipse XDB-C18(250 mm×4.6 mm,5 μm)was used with a mobile phase of CH3CN-H2O,flow rate:1 mL·min-1,column temperature:35 ℃,:spray temperature 39 ℃,drift tube temperature:55 ℃,air pressure:172.4 kPa,gain:200.Results:The calibration curves of astragaloside Ⅰ,astragaloside Ⅱ and astragaloside Ⅳ were in good linearity over the ranges of 0.8312–8.312 μg(r=0.9996),2.544–16.536 μg(r=0.9990) and 0.328–3.28 μg(r=0.9992) respectively;The average recoveries were 99.1%,98.0% and 98.6% with RSD of 3.9%,2.8% and 2.8%,respectively.Conclusion:The method is simple,rapid,sensitive and reproducible for determination of astragalosides in Radix Astragali.and can be used a basis for finding better habitat of Radix Astragali.
