Effect of nourishing yin improving blood circulation and detoxification ultrafine Chinese medicine on the expression of prothrombin mRNA and PAR-1 mRNA in cerebral infarction rats

Abstract

Objective: To observe the effect of nourishing yin improving blood circulation and detoxification ultrafine Chinese medicine prescription on the expression of prothrombim mRNA and protease-activated receptor-1(PAR-1) mRNA at brain tissue of focal cerebral infarction rats.Methods: Twenty-four rats were randomly divided into 4 groups: the sham-operated group,model group,Ziyin Huoxue Jiedu Ultrafine Chinese Medicine group(ZUG) and argatroban group(ABG).Each group was divided into two time phase point of 1st day,3rd day.The model was reproduced by occluded middle cerebral artery(MCAO).The expression of prothrombin mRNA and PAR-1 mRNA of brain tissue were detected by reverse transcription-polymerase chain reaction(RT-PCR) technique.Results: The low level of prothrombin mRNA and PAR-1 mRNA were detected in the sham-operated group.The expression of prothrombin mRNA and PAR-1 mRNA of brain tissue in model group,ZUG and ABG was significantly higher than that in the sham-operated group(P0.01),and the expression of prothrombin mRNA and PAR-1 mRNA of brain tissue in ZUG and ATB was significantly lower than that in the model group(P0.01);Compared with argatroban group,the expression of prothrombin mRNA and PAR-1 mRNA in ZUG increased obviously at first day(P0.05),but no difference on PAR-1 mRNA at third day.Conclusion: nourishing yin improving blood circulation and detoxification ultrafine Chinese medicine and argatroban are effective to restrain the expression of prothrombin mRNA and PAR-1 mRNA of brain tissue.It may be one of cerebral protective effects of nourishing yin improving blood circulation and detoxification ultrafine Chinese medicine that reduce thrombin toxic injury.
