Abstract
Objective To study the effects of benzimidazole on the expression of vimentin and microtubule proteins in rat testicular Sertoli cells in vitro culture. Methods Sertoli cells were harvested from rat testes aged 18 ~ 20 days,and then purified by Tris-Hcl low permeability treatment. The growth rates of Sertoli cells were determined with MTT assay in the negative control group,the solvent control group and benzimidazole dose groups. The expression of vimentin and microtubule proteins was detected by immunocytochemistry method. Results The proliferation of Sertoli cells was significantly inhibited in 10 and 100 μg /ml benzimidazole groups in 24,48 and 72 h. The expression of vimentin and microtubule proteins in Sertoli cells in 10 and 100 μg / ml benzimidazole groups was significantly decreased. Conclusion Benzimidazole has obvious toxic action to rat Sertoli cells in vitro culture,inhibites the proliferation of Sertoli cells,affects the expression of vimentin and α / β-tubulin in Sertoli cells.
