Abstract
Objective:To develop an HPLC method for determination of five isoflavones including puerarin,daidzin,genistin,glycitein and genistein,in foods made of pueraria simultaneously.Methods:A Cosmosil C18-AR(250 mm×4.6 mm,5 μm)was applied;a gradient mobile phase composing of methanol and water was used,and the flow rate was 1.0 ml/min;detective wavelength was set at 250 nm.Results:The results were obtained that quantitative linear range of five isoflavones was 0.035 μg~1.76 μg(r=1.0000),0.051 μg~0.20 μg(r=0.9995),0.045 μg~0.18 μg(r=0.9992),0.047 μg~0.18 μg(r=0.9998),0.055 μg~0.21 μg(r=0.9997),The average recoveries(n=6) were 98.58%,110.38%,98.58%,100.04% and 100.67%,respectively.Conclusion:A specific,sensitive,simple and reproducible HPLC method has been established for the determination of the five isoflavones in foods made of pueraria.
