Abstract
Objective:To develop an HPLC method for determination of six isoflavones, daidzin, glycitin, genistin, daidzein, glycitein, and genistein,in functional foods simultaneously. Methods:An Agilent Zorbax SB-C_(18) column (4.6 mm×150 mm,5 μm)was applied;a gradient mobile phase composing of acetonitrile and 0.2% formic acid was used, and the flow rate was 1.0 mL·min~(-1);detective wavelength was set at 255 nm.Results:The results were obtained that quantitative linear range of six isoflavones was 0.42-3.15,0.20-2.25,0.41-3.06,0.15-1.14, 0.051-0.38,and 0.21-1.55 μg respectively, and the average recoveries (n=5) were 95.86%,96.87%, 96.85 %,96.63 %,96.48 %,and 96.67 % respectively. Conclusion:A specific, sensitive, simple and reproducible HPLC method has been established for the determination of the six isoflavones in functional foods.
