Abstract
Five methods,i.e. CTAB,high saltlow pH,basic method,high salt precipitation, improved SDS method, were used to extract total DNA from the young leaves of grape. The resulted DNA samples of five methods were tested qualitatively and quantitatively using the value of A260/A280 、agarose gel electrophoresis and RAPD. The results show that high salt precipitation and high saltlow pH methods are better as a whole. The purity of extracted DNA by improve SDS is the best. So we should select the proper method according to the objetive in the study.
