Abstract
Three methods(CTAB method,SDS method and improved CTAB method) for the extraction of genomic DNA in rice seedling were compared.The result showed that the improved CTAB method was the best method for the extraction of DNA among three methods,and the DNA extracted by this method had higher yield,better quality,and could be used for RAPD-PCR reaction.A number of factors,which affected the results of RAPD-PCR reaction,such as quantity of template DNA,concentration of Mg2+,dNPT and Taq DNA polymerase,were studied.The optimized RAPD analysis system of rice was established.Several clear and stable bands with polymorphism were obtained.
