Optimization of Solid-Liquid Extraction of Polysaccharides from Mulberry Leaves using Orthogonal Methodology

Abstract

This work was focused on optimization of solid-liquid extraction of polysaccharides from mulberry leaves without flavonoids by orthogonal methodology.Single factor test showed that extraction temperature,time,ratio of raw material to water(g:mL),initial pH of extraction solution,and extracting number of time had different effect on the yield of mulberry polysaccharides(MP).The orthogonal results indicated the optimized conditions of extraction were as follows: extraction temperature 100 ℃,extraction time 4 h,ratio of raw material to water 1:30,initial pH 7,and extracting 2 times.Validation experiments suggested that the MP yield reached up to 2.74% under these optimized conditions.Additionally,this process was stable and easy to be industrialized.
