Abstract

Objective:To study the chang of the effective contents in Paeonia lactiflora Pall.befor and after the radiation of Co-60 rays. Methods:The HPLC was adopted. The separation was performed on a Switzerland SPHERIGEL ODS C18(150 mm×4.6mm,5 μm)column with methanol-water as mobile phase in gradient elution(0-10min,15%→20% methanol;10-30 min,20%→30% methanol). The flow rate was 1.0 mL·min-1,the detection wavelength was set at 260.5nm,and the column temperature was maintaned at 25 ℃.Analysis time was 30 minutes..Results:The effective contents in Paeonia lactiflora had been reduced nearly 20% after the radiation of Co-60 rays. Conclusion:By investigating with systematic methodology, the techcique is convenient, stable and repeatable. It can be regarded as a standard in evaluating the content of paeoniflorin in Paeonia lactiflora before and after the irradiation of Co-60 rays.It also is a reference to the experiments of irradiating Chinese medical herbs.
