The anti-tumor effects of CIK against ovarian cancer cell lines SKOV-3 in vitro

Abstract

Objective To investigate the anti-tumor effects of cytokine-induced kill cells(CIK) against ovarian cancer cell lines SKOV-3 in vitro. Methods Peripheral blood mononuclear cells(PBMC) of healthy human were stimulated by different cytokines, and were induced into CIK cells.The phenotype of CIK cells were analyzed by a flow cytometer. CIK cells cultured for 14 days and logarithmic phase SKOV-3 lines were taken as effect cells and target cells respectively. Effect cells were co-cultured with target cells at different ratio.The antitumor effect of CIK cells against SKOV-3 lines was measured by MTT. SKOV-3 cells were cultured with the CIK cells culturing supernatant for 24 and 48 hours and the SKOV-3 cells apoptosis was analyzed by a flow cytometer. Results The CD3+CD56+double positive cells in the ratio was up to(31.6±5.5)% after 14 days culture. The killing activity increased with the cultured time prolong and the effector-target ratio increase. The maximum killing efficiency was about(74.74±6.87)% after co-culturing 48 hours with the effector-target ratio40∶1. The CIK cells supernatant can induce apoptosis of SKOV-3 cells.The apoptosis rates were(12.30±1.47) %and(27.13±2.03)% respectively after co-culturing 24 and 48 hours. Conclusion CIK cells have strong anti-cancer activity against SKOV-3 cells in vitro by inducing apoptosis of SKOV-3 cells.
