Effect of Resveratrol on Human CIK Cells

Abstract

Objective To investigate the effect of resveratrol on human CIK cells and in vitro cytotoxicity.Methods CIK cells that expressed CD3+CD56+ more than 15% were obtained through routine culture method in vitro.The different concentrations of resveratrol were used to induce human CIK cells.After 48 hours:the proliferation of human CIK cells was measured by MTT method,the expression of perforin and granzyme B in human CIK cells were detected by flow cytometry(FCM),and the cytotoxic activity of CIK cells against gastric cancer cell line SGC-7901 were measured by using LDH assays.Results 0-1.6 μmol/L resveratrol promoted the growth of CIK cells and increased the expression of perforin and granzyme B in CIK cells.But the CIK cells growth were inhibited by high concentration of resveratrol(more than 3.1 μmol/L),and inhibition rate were increased with the concentration.Conclusion The resveratrol at a given concentration can promote the growth of CIK cells and improved the killing activity of CIK.
