Abstract

Objective To study the anti-respiratory syncytial virus(RSV) effect of an compound(RG3-1) from radix gentianae in vitro. Methods RG3-1 was extracted from radix gentianae by hot water, dichloromethane, and column chromatography. The cytotoxicity and antiviral action of RG3-1 were tested by cytopathic effect(CPE) inhibition assay, and ribavirin as the positive control drug. Results It showed good inhibitory effect against RSV with the medium cytotoxicity concentration(TC50) 5.29mg/mL,the medium effective concentration(EC50) 262.95μg/mL,and the treatment index(TI) 20.12.RG3-1 was effective antiviral agent for RSV in a concentration-dependent manner.RG3-1 also displayed direct inactivation on RSV.It also exhibited obvious inhibition on RSV replications when RG3-1 was added in 10h after infection(P0.01).Conclusion RG3-1 can significantly inhibit RSV replications in HeLa cells.
