Enzymatic Extraction of Polysaccharide from Angelica

Abstract

Objective: Enzymatic extraction technology of polysaccharide from Angelica and its antitumor activity were studied.Method: The effects of enzyme dosage,enzymolysis temperature,enzymolysis time,enzymolysis pH value on the extraction rate of polysaccharide were studied by single-factor test.Then,the orthogonal experiment was performed to screen the optimum extraction technology.Antitumor activity of Angelica polysaccharide was evaluated by MTT method in hela cells.Result: The factors influencing the extraction process were in the order:enzyme dosageenzymolysis timeenzymolysis pH valueenzymolysis temperature,and the optimum extracting technology was as follows: enzyme dosage 0.4mg/ml,enzymolysis temperature 60℃,enzymolysis time 4h and enzymolysis pH value 5.The polysaccharide extraction rate was 150.34mg/g under these conditions.The results of MTT method confirmed that Angelica polysaccharide had some antitumor activity.Conclusion: The enzymatic extraction technology is simple with low cost and provides a scientific basis for the further development of Angelica polysaccharide.
