Abstract
Objective: To observe the stability of type 1 diabetes rat model(T1DM) induced by streptozotocin(STZ),and to explore standards of successful modeling. Methods: T1DM model was established by intraperitoneal injection of STZ with a dose of 50 mg/kg,and rats in control group were given the same volume of pH 4.2,0.1 mol/L citric acid-citric sodium buffer solution instead. The fasting blood glucose of each group was detected respectively in 72 hours,1 week,2 weeks,3 weeks,4 weeks,5 weeks,6 weeks,7 weeks, and 8 weeks after injection of STZ.Results: According to common standards of successful model that the fasting blood glucose greater than or equal to 16.7 mmol/L after intraperitoneal injection of STZ for 72 h,the modeling successful rates of our T1DMs after injection for different time durations were 100%,80%,75%,70%,70%,70%,70%,70% and 70% respectively. Conclusions: Based on the current standards,30% T1DM model rats are failure actually.Fasting blood glucose ≥16.7 mmol/L kept for 3 weeks after injection of STZ as the standard for succesful modeling is proposed here.
