Abstract
In order to establish a rat model of type 2 diabetes mellitus(T2DM) induced by streptozotocin(STZ) in combination with high-sugar and high-fat diets,30 healthy male SD rats were randomly divided into the T2DM model group(20 rats) and the blank group(10 rats).STZ(30 mg/kg)was injected into rats in the T2DM model group.Rats in the T2DM model group were fed with high-sugar and high-fat diets for 4 weeks.The fasting blood glucose(BG),blood triglyceride(TG),blood cholesterol(TC),body weight,fasting insulin(INS),glucose dependent insulinotropic releasing peptide(GIP),insulin-like growth factor-1(IGF-1),glucagon like peptide-1(GLP-1) were detected in first week,fourth week and after first week,fourth week,eighth week,twelfth week after STZ injection.The sections of pancreas were detected by histopathological examination.The success rate was 75%.Values of TG,TC in T2DM model group increased significantly than those in blank group(P0.01)after 4 weeks.Values of BG,TG,TC in T2DM model group increased significantly than those in blank group(P0.01)1,4,8,12 weeks after injection of STZ;ISI decreased significantly than those in blank group,P0.01;values of GLP-1,IGF-1 in T2DM model group decreased significantly than those in blank group(P0.01)1,4,8 weeks after injection of STZ;values of GLP-1,IGF-1,GIP in T2DM model group decreased significantly than those in blank group(P0.01)12 weeks after injection of STZ.The method of inducing T2DM by high-sugar and high-fat diet with STZ injection was effective.GIP,IGF-1,GLP-1 involved in the pathogenesis of type 2 diabetes mellitus.
