Simultaneous determination of chlorogenic acid,rutin,hyperoside and quercetin in Fructus Chaenomelis by HPLC

Abstract

Objective To establish an HPLC method for the simultaneous determination of chlorogenic acid,rutin,hyperoside and quercetin in Fructus Chaenomelis.Methods Ahigh-performance liquid chromatography equipped a Phenomenex luna C18(250 mm×4.6 mm,5 μm) column with UV detection used.The mobile phase consisited of 0.6% aqueous phosphoric acid(A) and acetonitrile(B) with gradient elution.The detection wavelength was set at 360 nm and the flow rate was 1.0 ml·min-1.Results The linear ranges of chlorogenic acid,rutin,hyperoside and quercetin were 0.035~0.56 g·L-1(r=0.997 8),0.003 2~0.051 2 g·L-1(r=0.999 3),0.004 25~0.068 g·L-1(r=0.999 7),0.003 5~0.056 g·L-1(r=0.999 7),respectively;the average recoveries were 102.9%(RSD=1.04%),99.3%(RSD=2.28%),99.2%(RSD=2.33%),99.5%(RSD=2.52%).Conclusion The proposed method is simple,accurate,and repeatable.which can be used for quality control of Fructus Chaenomelis.
