The Effect of CGRP and NGF on Expression of Bcl-2 in Temporary Ischemia cerebral Tissue in Rats

Abstract
Objective: To observe the effect of calcitonin gene related peptide(CGRP)and Nerve growth Factor (NGF) on expression of Bcl-2 protein in ischemic brain tissue and probe into the protective effect of it on ischemic neuron. Methods: The model of whole cerebral ischemia/reperfusion in rats were induced by means of carotid artery migatie pressure shunt(CANPS); CGRP and NGF was injected into internal carotid at the beginning of reperfusion; The expression of Bcl-2 in brain tissue was detected by method of immunohistochemistry. Results: Compared with ischemic group(24.88±2.88 and 2.95±0.34), the percentage of Bcl-2 immunoreaction positive cells in cortex and hippocampus CA1 in CGRP group increased significantly(70.83±8.95 and 4.17±0.34 respctively), (P0.01); The percentages of Bcl-2 immunoreaction positive cells in cortex and hippocampus CA1 in CGRP plus NGF group were 83.23±5.97 and 8.03±1.56 respectively, much higher than that of in CGRP group (P0.01). Conclusion: CGRP might upregulate the expression of Bcl-2 in brain tissue after wholecerebral ischemia in rate, which might be contributed to their protective effects on ischemia neuron. The combined usage of CGRP and NGF might enhance such effects.
