Relationship between K-ras mutation in pancreatic juice as well as serum CA19-9 level and recurrence of pancreatic carcinoma

Abstract

Background and purpose:K-ras is the gene associated with pancreatic carcinoma.And CA19-9 levels are considered valuable for the diaguosis of panceatic carcinoma.So we studied whether K-ras point mutation in pancreatic juice and serum CA 19-9 level are associated with recurrence of pancreatic carcinoma after surgery.Methods:K-ras point mutation was detected using the polymerase chain reaction and restriction fragment-length polymorphism(PCR-RFLP) in pure pancreatic juice(PPJ),which were collected from the pancreatic togarphy(ERCP).And serum CA19-9 levels were examined in 32 patients with pancreatic carcinoma.The relationships among K-ras and serum CA19-9 level and recurrence rate were analyzed.Results:① 56.3% of K-ras point mutation in PPJ was positively detected,which is significantly correlated with the recurrence of tumor(P0.05).② The recurrent rate is higher in the patient with positive K-ras point mutation in PPJ and high serum CA19-9 level than the in those with low serum CA19-9 level(69.2 vs 20.0%,P0.05).Conclusions:The combined detection on K-ras point mutation in PPJ and serum CA19-9 level can be useful to predict the possibility of recurrence after surgery.The analysis of gene profile in PPI is valuable for the predictive accuracy.
