Abstract
Objective: To observe the protective effect of (Daphnetin) on myocardial ischemia/reperfusion injury in rats.Methods:Rat's ischemia/reperfusion injury model was established by ligating left anterior descending coronary artery for 32 min followed by reperfusion for 120 min.48 male rats were randomly divided into 4 groups,the sham operation group,the model group,The two treated groups.The two treated groups were treated with Daphnetin respectively by given the drugs 10 min by way of sublingual vein before ischemia reperfusion,and to the sham operation group and the model group,saline was given instead.The infarction area,pathologic change cardial tissue(under light microscopy),activity of creatine phosphokinase(CK) and lactate dehydrogenase(LDH) in serum,content of malondialdehyde(MDA) and superoxide dismutase(SOD) in cardial tissue were observed to evaluate the protective effect of treatment.Results The area of acute mycardial infarction was lesser,activity of LDH and CK were lower in the two treated groups than those in the model group.Content of SOD was signifimntly higher and that of MDA was markedly lower in the former two than those in the model group.And all the changes mentioned above show Dose-dependent effect,To the Daphnetin(30mg/kg)though it showed a little effect but had no significant difference.Light microscopic examination showed that the necrotic degeneration and pathologic changes of myocardiocytes in the treated groups were a little milder than that of the model group.Conclusion Daphnetin have protective effects on myocardium against ichemia reperfusion injury.
