Protective effect of tetramethylprazine on myocardium in acute myocardial infarction rats

Abstract

Objective To investigate the protective effect of tetramethylprazine on myocardium in experimental myocardial infarction rats and its mechanism.Methods A total of 40 Wistar rats were involved in this study.The living rats were randomly divided into three groups,which were control group(n=10),acute myocardial infarction group(n=15) and tetramethylprazine group(n=15).Acute myocardial infarction model was established by ligating left anterior descending artery of rats.Rats in tetramethylprazine group were injected with tetramethylprazine by means of lumbar injection at 120 mg/kg perday and lasted for four weeks.The control group experienced the same procedure with acute myocardial infarction group in operation,but with no ligation of coronary artery.Four weeks after operation,doppler echocardiography and serological examination were used to test cardiac function in each group.All rats were killed to record their body weight and heart weight.Results The left ventricular end-diastolic dimension(LVDd),left ventricular end-systolic dimension(LVDs) and E/A ratio of acute myocardial infarction group were higher than those of control group,while ejection faction(EF) and fractional shortening(FS) were lower than those of control group(all P0.05).LVDd,LVDs and E/A ratio in tetramethylprazine group were lower than those of acute myocardial infarction group,while EF and FS were higher than acute myocardial infarction group(all P0.05).Vascular endothelial growth factor(VEGF) and superoxide dismutase(SOD) of acute myocardial infarction group were lower than those of tetramethylprazine group(all P0.05).VEGF,LDH were closely related with EF both in acute myocardial infarction group and tetramethylprazine group.Conclusion Tetramethylprazine have certain effect on protecting myocardium in rats with acute myocardial infarction by increasing the contents of VEGF in serum and the activity of SOD.
