Effect of celecoxib on the expression of IL-8 and MCP-1 in peripheral blood of rats with acute lung injury

Abstract

Objective To investigate the effects of celecoxib on the expressions of peripheral blood chemokines-interlukine 8( IL-8)and monocyte chemoattractant protein( MCP-1)in rats with acute lung injury induced by lipopolysaccharide( LPS).Methods Forty-five rats were randomly divided into three groups:control group,LPS group and celecoxib treatment group,with 15 rats in each group.The rats in control group were given 0.9% sodium chloride solution,the animal models with acute lung injury were established by intravenous injection LPS( 5 mg/kg)and the rats in celecoxib treatment group were given celecoxib( 20mg/kg)at 30min after LPS induction,then the rats were sacrificed after 3h.The expression levels of IL-8 and MCP-1 in peripheral blood of rats were detected by ELISA.Results There were significant differences in the expression levels of IL-8 and MCP-1 between LPS group and control group( P 0.05).There were significant differences in the expression levels of IL-8 and MCP-1 between celecoxib treatment group and LPS group( P 0.05).Conclusion Celecoxib can improve the reaction extent of rats to acute lung injury by inhibiting the expression levels of IL-8 and MCP-1.
