Abstract
Biological characteristics of Phyllosticta citrichinaensis was studied through observation of mycelial growth and spore germination in in vitro culture under different media, temperature,pH value and light. Results showed that adding Guanxi-miyou leaf juice into the media could stimulate the growth of mycelial. The temperature for mycelial growing was 10~35℃,with the suitable temperature from 25℃ to 30 ℃,and optimum temperature at 28℃.At 40℃,the mycelial stopped growing.Mycelial could grow in a pH range of 3~9,with the optimum pH at 5~6.The edge of colony turned into yellow after 14 days culture on pH 7~9 oat meal agar(OA) medium. The germination rate of conidium reached maximum of 45%at 25℃ and pH 4.The 12-hour light/dark treatment could stimulate mycelial growth. The lethal temperatures for mycelial growth and spore germination were 55℃ for 10 minutes and 50℃ for 10 minutes, respectively.
