Abstract

The effects of temperatures,pH values,illumination,carbon sources,nitrogen sources and different media on the mycelial growth,sporulation and conidial germination of Colletotrichum higginsianum Sacc.were tested.The results showed that the temperature for the mycelial growth and sporulation was from 8 to 35 ℃,with an optimum of 25 ℃;within 10?35 ℃,the conidia could germinate,with an optimum of 25 ℃;The lethal temperature of conidia was 53 ℃,10 min or 55 ℃,5 min.The pH values for the mycelial growth and sporulation were 3 0?10 0,with an optimum of pH 6 0.Illumination had no effect on the linear growth of mycelium,but continuous illumination could increase the mycelial dry weight and the amount of conidia.In the tested nine carbon sources,mycelial growth was the fastest on the media using mannitol,maltose and glucose as carbon sources,respectively;mycelial dry weight was the highest on the medium using glucose as carbon source;and the amount of conidia was the largest on the medium using maltose as carbon source.In the tested nine nitrogen sources,mycelial growth was the fastest on the medium using glycine as nitrogen source;mycelial dry weight was the highest on the medium using potassium nitrate as nitrogen source;and the amount of conidia was the largest on the medium using urea as nitrogen source.In the tested eleven media,mycelial growth was the fastest on the media of PDA and the flowering Chinese cabbage juice PDA,respectively;and the amount of conidia was the largest on the medium of corn meal agar.
