Abstract

Objectives To observe the inducing effects of angiotensinⅡ(AngⅡ)on the senescent of human umbilical vein endothelial cells(HUVECs)and the intervention effect of Kudiezi Injection.Methods HUVECs were cultured in vitro and intervened by AngⅡ,then were divided into three groups:control group,AngⅡgroup (stimulated with AngⅡ10-6 mol/L for 48 h),and Kudiezi Injection group(pre-administered by 10%Kudiezi Injection before treated by AngⅡ).The cell viability was observed by CCK-8,two ageing markers of ageing associated β-galactosidase activity and cell proliferation activity were adopted as main indexes.β-galactosidase activity was measured by immunocytochemical method and cell proliferation activity was measured by flowcytometry.The expression of caveolin-1 was determined by Western-bloting.Results As compared with control group,the positive rates forβ-gal staining was significantly increased in AngⅡgroup,and showed cell cycle arrest at G0/G1 phase with decreased S-phase cell percentage and cell viability.The expressions of caveolin-1 protein were significantly increased in AngⅡgroup(P0.01),which indicated that HUVECs senescence were successfully induced by AngⅡ.The above changes were smaller in Kudiezi Injection group than in AngⅡgroup.Conclusions Kudiezi Injection can delay AngⅡ-induced aging of HUVECs possibly by down-regulating the expression of caveolin-1 protein.
