Abstract
Objective To investigate the combined effects of kallikrein and valsartan(Val) on reversal of left ventricular remodeling,angiotensin Ⅱ(Ang Ⅱ) and serum nitric oxide(NO) level in spontaneously hypertensive rats(SHRs).Methods Twenty-four 15 week-old SHRs and 8 fifteen-week male Wistar-Kyoto rats(WKYs) were divided randomly into SHR group,pancreatic kininogenase(PK) group,Val+PK group and WKY group to conduct experiment lasting for 4 weeks.Ang Ⅱlevels were detected by radioimmunology;plasma NO contents were determined by chemical method;systolic blood pressure(SBP),left ventricular mass index(LVMI), collagen volume fraction(CVF) and perivascular collagen area(PVCA) were detected by routine method.Results In SHR group,SBP,LVMI,CVF,PVCA and Ang Ⅱ were higher,serum NO level was decreased obviously than those of WKY group(P0.01).In each treatment group,serum NO level increased(P0.01) obviously,other indexes obviously reduced(P0.01) than those in SHR control group significantly(P0.01).Plasma Ang Ⅱ levels increased significantly(P0.01),myocardium Ang Ⅱ levels decreased(P0.01) significantly in Val+PK group.But there was no obvious change in Ang Ⅱ levels in PK group.Conclusions Val and PK all could effectively ameliorate left ventricular remodeling of hypertension,especially combined Val and PK.
