Abstract
Objective To explore the relationship between the expression of survivin and IGF-1 and neuronal of apoptsis after cerebral ischemic reperfusion injury in rats.Methods The healthy adult of male Wister rats (80) were divided into Survivin group and IGF-1 group,per group (40),and they divided andomly into control group and sham-operation group and in ischemic 1h reperfusion 2h,6h,12h,24h,48h,3d,7d,14d group(n=4).The models with cerebral ischemic reperfusion were induced by intraluminal middle cerebral artery occlusion (MCAO) with a nylon monofilament suture. And using immunohistochemistry technique detecte the condition of the expressionof of Survivin protein and IGF-I protein in apoptosis of neurons.Results Survivin group: Showing the expression of background of Survivin positive neuron in hippocampus and cortical area and striatal area on sham-operation group.The expression is increased clearly from ischemic reperfusion in 2h,reached peak in 48h,and is best lower in 14d.It compare with sham-operation is significancely difference(P0.05).IGF-1 group:Showing the expression of background of IGF-1 positive neuron in hippocampus and cortical area and striatal area.The expression of IGF-1 is clearly increased in ischemic reperfusion in 2h,reached peak in 48h,and is constant in 3-14d. It compare with normal control group and sham-operation is significancely difference(P0.05).Conclusion The expression of endogenous survivin and IGF-I might inhibited apoptsis of neuron.
