Abstract
Objective: To establish HeLa cell line with stable expression of human autoimmune regulator. Methods: Eukaryotic expression vector plasmid containing AIRE cDNA was transfected into HeLa cells with FuGENE6 Transfection Reagent. The positive cell clones had been obtained after selected by G418. The stable transfection and expression of AIRE in HeLa cells were determined by the fluorescence microscope, FACS and Western blotting. Results: Stable expression of AIRE was confirmed by fluorescence microscope, FACS and western blotting analysis. Conclusion: With FuGENE6 transfection reagent, AIRE gene was stably expressed in HeLa cells that would be helpful for further study on AIRE gene.
