Abstract

Five breeds of geese(Yangzhou goose,Sichuan White goose,Wanxi White goose,Huoyan goose and Zhedong White goose)with 30 male and 30 female each were used in this experiment.One pair of primer was designed for amplification of intron fragment based on adiponectin gene sequence of goose,and then single nu-cleotide polymorphism(SNP) and genetic polymorphism in different breeds were analyzed by PCR-SSCP method.As a result,two single nucleotide polymorphism mutations(A16G and T26C) were found in adiponectin gene and three genotypes(AA,AB,and BB) were detected in the five breeds.Except for that between Huoyan goose and Wanxi White goose,and Huoyan goose and Zhedong White goose,the distribution of three genotypes was not significantly different between every two breeds.The heterozygosity and PIC(polymorphism information content) were highest in Huoyan goose,but lowest in Wanxi White goose.Every breed′ s PIC was in the middle level.The results showed that the adiponectin gene has high polymorphisms in goose breeds,and could be used as a candi-date gene for analyzing the correlation between its polymorphism and adipo traits in goose.
