Polymorphism analysis on exon 2 of adiponetin gene in different breeds of geese

Abstract

To search single nucleotide polymorphisms(SNPs) in exon of adiponectin gene in domestic goose,and to identify SNP location in goose breeds,then to analyze genetic parameter of goose populations,two pairs of primers were designed to detect SNPs of exon 2 on adiponetin gene in five breeds of goose by polymerase chain reaction-single-strand conformation polymorphism(PCR-SSCP).The results showed that one SNP(A→G) was found in the 351 bp of exon 2,but it didn't cause the change of amino acid sequence.The allele frequencies for X and Y of Huoyan goose were the closest,being 0.566 7and 0.433 3,respectively.However,the frequencies of allele Y were significantly higher than those of allele X in other four goose breeds.The genotypes of Huoyan goose differed significantly from those of other goose breeds(P 0.01).The genotypes of Zhedong white goose was significantly different from Taihu goose and Sichuan white goose(P 0.05).Except for Zhedong white goose,the polymorphism information contents(PICs) of other breeds were moderate.The adiponectin gene,due to its high polymorphism in goose breeds,could be employed as a candidate gene in goose breeding.
