Abstract
In this experiment,one pair of primers in exon 2 based on sequence of GH gene of goose and duck was designed to detect the single nucleotide polymorphisms(SNPs) by the technique of single strand conformation polymorphism(SSCP) and the genetic polymorphism was analyzed,taken six goose breeds(Taihu goose,Yanzhou goose,Huoyan goose,Zhedong White goose,Wanxi White goose and Sichuan White goose)as material.Two single nucleotide polymorphism sites were discovered in exon 2.T39C variation did not cause the change of amino acids,but T74C variation resulted in the amino acid variation of V25A.Ten genotypes(AA,AB,AC,AD,BB,BC,BD,CC,CD,DD) were detected in the six breeds.The result of χ2 analysis showed that the distribution of ten genotypes was significant different among breeds(P0.01).All breeds were in accordance with the Hardy-Weinberg equilibrium.The heterozygosity,effective number of alleles and polymorphism information content(PIC),was lowest in Taihu goose,but highest in Zhedong White goose.Except Taihu goose′s PIC was in the middle level,the other′s PIC were in high level.The results showed that the GH gene has high polymorphisms in different goose breeds,and likely to be a candidate gene for analyzing the correlation between its polymorphism and production traits in goose.
