Effect of atorvastatin on the expression of IL-8 and CTGF in border zone myocardium after acute myocardial infarction in rats

Abstract
Objective:To investigate the effect of atorvastatin on the inhibition of ventricular remodeling in border zone myocardium.Method:Forty-five male AMI rats induced by left anterior descending branch ligation were randomly divided into AMI group,high dose atorvastatin treatment group and low dose atorvastatin treatment group.Another ten rats were taken as the sham operation group.The high and low dose group were respectively treated with volume of 20mg·kg-1·d-1 and 10mg·kg-1·d-1 atorvastatin for 4weeks by gavage,whereas the sham operation group was not given any treatment.At 4th week,the left ventricle free wall tissue in the sham operation group and the border zone myocardium in the rest of rats were preserved.Myocardial collagen staining and the expression of myocardial CTGF and IL-8were determined.Result:Compared to the sham operation group, there were higher expressions of myocardial IL-8,CTGF and blue-stained collagen in the border zone in the AMI group.Compared to the the AMI group,low dose atorvastatin treatment reduced the expressions of myocardial blue-stained collagen,IL-8and CTGF(P0.05).Compared to the the low dose atorvastatin group,high dose atorvastatin treatment reduced the expressions of myocardial blue-stained collagen,IL-8and CTGF(P0.05). Conclusion:Atorvastatin may inhibit ventricular remodeling in the border zone after AMI in rats by inhibiting the over-expression of myocardial IL-8and CTGF.The effects depend on the dose of atorvastatin.
