Extraction and Saponification Technology of Lutein from Marigold

Abstract

In this paper,Extraction and Saponification technology of Lutein esters in Tagetes erecta L.particle were investigated systemically.The mixed solvent of THF and Alc is main extraction solvent.The affecting factors such as the ratio of THF and Alc,reaction time,the ratio of reaction solution to material(mL/g),the addition of water,the addition of KOH in reaction solution and reaction temperature were investigate in singe factor experiments.An orthogonal test table L9(34) was used to optimized the affecting factors on the base of singe factor experiments.The content of free Lutein in the end product was determined by HPLC.The results showed that the optimal Extraction and Saponification parameter was:1.6g KOH,4h,the ratio of 1∶20(mL/g),ratio of THF and Alc is 2∶8.Under this condition,the yield of Free Lutein reach 1.14% and the content of Free Lutein reach 10.32 % in extracta sicca.
