Screen and Mutation of a Degradation Bacterium for Sesame Meal and Its Molecular Identification

Abstract

A bacteria strain LQ,which can degrade the crude protein of sesame meal,was screened from the nature.In addition,its 16S rRNA gene sequence was determined,its molecular phylogenetic tree was constructed,and its homology with corresponding sequence of related bacteria was analyzed.The results showed that the length of the 16S rRNA gene sequence of LQ was 1 447 bp;natural clustering the 16S rRNA gene sequence of LQ with the 16S rRNA gene sequence of bacteria(such as Bacillus Cohn,etc.),the results indicated that among the retrieved sequences of Bacillus,the homology between LQ and Bacillus amyloliquefaciens reached 99%,therefore,the LQ is identified as Bacillus amyloliquefaciens.After the LQ strain was mutated by UV mutagenesis,its activity of protease reached 587.3 U/mL.
