Clinical study of oxaliplatin in combination with 5-fluorouracil plus calcium folinate on advanced gastric cancer

Abstract

Objective To evaluate the clinical efficacy and adverse effects of oxaliplatin combined with 5-flu-orouracil(5-FU)/calcium folinate(CF) in the treatment of advanced gastric cancer. Methods Oxaliplatin 100 mg/m2 ivgtt 2hr d1,CF 400 mg/m2 ivgtt d1,5-FU 400 mg/m2 iv bolus d1,5-FU 2600 mg/m2,contimuous intravenous drip(CIV) 46 h,and repeated every 2 weeks.Efficacy was evaluated at 4 cycles (8 weeks). Results Forty-six patients were evaluable for clinical response.Five patients achieved complete response(CR)(11%),18 partial response(PR)(39%),16 stable of disease(SD)(35%),7 progress of disease(PD)(15%),overall response rate(CR+PR) was 50%.Response rates for 1st line and 2nd line patients were 58% and 40%,respectively.Median response time was 5 months and median survival time was 9 months.The main adverse effects were neural toxicity.Nausea/vomiting,leukopenia and thrombocytopenia were mild. Conclusion Oxaliplatin combined with 5-FU/CF is a high response regimen for advanced gastric cancer with mild toxicity,which can be practiced safely.
