Abstract

The regeneration system of four cultivated potatoes in vitro was studied with leaves as explants,and the results showed that the optimal medium for callus induction from leaf discs was MS+6-BA 2.5 mg·L~(-1)+NAA 0.2 mg·L~(-1) for “Dongnong 303” and “E No.1”,MS+6-BA 2.0 mg·L~(-1)+NAA 0.1 mg·L~(-1) for “Favorite” and MS+6-BA 2.0 mg·L~(-1)+NAA 0.2 mg·L~(-1) for “Sharpdi”.The optimal medium for adventitious bud differentiation from leaf-derived calluses was found to be MS+6-BA 2.5 mg·L~(-1)+GA_3 5.0 mg·L~(-1) for “Favorite” and “E No.1,MS+6-BA 1.0 mg·L~(-1)+IAA 0.1 mg·L~(-1)+GA_3 2.5 mg·L~(-1) for “Dongnong 303”,MS+6-BA 2.5 mg·L~(-1)+IAA 0.5 mg·L~(-1)+GA_3 2.5 mg·L~(-1) for “Sharpdi”.Adventitious bud differentiation rate of “Dongnong 303”,“Sharpdi”,“Favorite” and “E No.1” reached 94.3%,100%,100% and 90% respectively.
