Study on in vitro regeneration technology from stem explants of four potato cultivars

Abstract

Four potato cultivars were regenerated in vitro from stem explants. The results showed that the optimal medium for callus induction was found to be MS + 6-BA 3. 0 mg/L + NAA 0. 01 mg/L for cv. Dongnong 303,cv. Shepody and cv. E No. 1, MS + 6-BA 3. 0 mg/L + NAA 0. 1 mg/L for cv. Favorite. The callus induction rate on those media reached up to 100%. The optimal medium for adventitious bud differentiation from stem callus was found to he MS+6-BA 2. 0 mg/L + NAA 0. 2 mg/L for cv. Favorite and cv. E No. 1,MS+6-BA 2. 0 mg/L + NAA 0. 1 mg/L for cv. Dongnong 303,and MS + 6-BA 2. 0 mg/L + NAA 0. 3 mg/L for cv. Shepody. The adventitious hud differentiation rate reached 96. 7%, 96. 7%, 83. 3% and 80. 0% for the four cultivars respectively,and the rooting rate was 100% for all cultivars. The survival rate of tube plantlets after transplantation reached 95%.
