Abstract

Objective To study the features of DNA damage induced by arsenic, fluoride and their combination in cultured SGC 7901 cells.Methods Single cell gel electrophoresis(SCGE) and Komet Image Analysis System (KIAS) were used to measure the DNA damage of the cells treated with 10 μmol/L sodium and/or sodium fluoride for 4 hours.Results ①The tail length and the tail moment of damaged DNA were significantly greater in the sodium arsenite group than that in the control group, the tailed cell percentage in the sodium fluoride group was significantly higher than that in the control group;②Such indicators as the tailed cell percentage, tail DNA,tail length and tail moment in the combined treatment group were significantly increased , however the interaction of DNA damage between arsenic and fluoride was not observed.Conclusions Both sodium arsenite and sodium fluoride could induce DNA damage, but their mechanism of inducing DNA damage may be different. And the DNA damage effects of sodium arsenite and sodium fluoride in the combination group were simply additive.
