Abstract

Objective:To study the genotoxicity of fluoride.Method:The effects of sodium fluoride on DNA damage and repair in isolated male mice testicular cells were observed with single cell gel electrophoresis test(Comet assay). Results:At the concentriation of 0 62mmol/L to 5mmol/L,sodium fluoride could induce the breakage of DNA single strand and resulted in comet cells with tail. Both the number of comet cells and the length of DNA migration increased with the increasing of concentration of sodium fluoride, showing the obvious dose effect relationship. In addition, the DNA damage could be repaired basically within two hours incubation. Conclusion:The research suggested that at certain concentrations, sodium fluoride could induce DNA damage in isolated male mice testicular cells and might be a kind of male reproductive toxicants.
