Screening Study for Anti-Depression Effect of Xiaoyao Powder Prescriptions and Their Separated Parts

Abstract
Objective:To investigate the anti-depression effect of Xiaoyao Powder prescriptions and their isolated components and to screen the best prescription and best part.Methods:Rats were treated orally with Xiaoyao Powder,Danzhi Xiaoyao Powder,Chaihu Shugan Powder or positive drug(venlafaxine) for two weeks.The FST was conducted 1 h after the last drug treatment to evaluate the anti-depression activity of Xiaoyao Powder prescriptions.Furthermore,the inhibition of the prescriptions and their six separated parts of different polarity on rat brain synaptosomal assimilate 5-hydroxytryptamine(5-HT),noradrenaline(NA) and dopamine(DA) were investigated by in vitro activity screening experiment.Results:After a two-week treatment,Xiaoyao Powder,Chaihu Shugan Powder and the venlafaxine reduced the immobility time significantly(compared to FST with control group,P0.05).In the condition that the final reaction concentration of the samples are 10 μg/mL,Xiaoyao Powder and Xiaoyao Powder separated parts X6,X7 could inhibit rat brain synaptosomal to assimilate 5-hydroxytryptamine(5-HT),noradrenaline(NA) and dopamine(DA) significantly.Conclusion:It is a suggestion that Xiaoyao Powder and its isolated parts of X6,X7 had the favorable anti-depression effect and they could be ideal components for screening novel antidepressant drugs.
