Effect of Rosiglitazone on Peripheral Leptin Resistance in Diet - Induced Obese Rats

Abstract

Objective To observe dynamic changes of leptin resistance and the relationship between leptin resistance and insulin resistance in diet-induced obese rats,and to observe the effect of rosiglitazone intervention on leptin resistance. Methods Thirty SPF level Wistar male rats of age 2-3 weeks were divided into 3 groups randomly: the normal control group, fat group and the rosiglitazone - treated fat group. After 20 weeks high fat diet feeding,fat models were established. The serum leptin,insulin concentration were detected in 10,20,26th week with radioimmunoassay. Results The serum leptin level in fat group was significantly higher than that in the control group and the rosiglitazone-treated fat group(F0.01,0.05).The serum leptin concentration was positively correlated with insulin levels(r =0.427 P0.05), triglyceride(r = 0.515 P0.01).Conclusions The serum leptin concentration may gradually increase with the worsening of insulin resistance . Rosiglitazone improves both insulin resistance and leptin resistance.
