Investigation of anchoring protein antibodies CD55/CD59 and RET on the diagnostic significance of anemic patients

Abstract

Objective By detecting the expression of membrane associated complement regulatory proteins(CRP),CD55 and CD59 on the peripheral blood cerythrocytes and neutrophils in several kinds of anemia patients,and analysing the percentages of reticulocyte to investigate the diagnostic value of them in anemic patients.Methods the percentages of CD55-/CD59-and reticulocyte(Ret%)of 30 normal subjects,25 cases of IDA,26 cases of MA,27 cases of AHA,27 cases of AA,21 cases AA-PNH and 30 cases of PNH patients were detected by luorescence-activated cell sorting(FACS) with PE and FITC fluorescence labeled anti-CD55 and anti-CD59 monoclonal antibodies,and by methylene blue intravital staining technology.Results The percentage of CD55-and CD59-cells in 30 normal subjects were less than 5%.The percentages of CD55-and CD59-on erythrocytes were in the range from 5% to 20%,on neutrophils were less than 5% in 44% of the patients with microcytic hypochromic anemia.The percentages of CD55-and CD59-on erythrocytes and neutrophils were less than 5% in patients with macrocytic anemia and autoimmune hemolytic anemia.The percentages of CD55-and CD59-on erythrocytes and neutrophils were in the range of 5% to 15% in about 33% patients with aplastic anemia,but the percentages in the patients with PNH and AA-PNH were more than 11%.compare to normal control,the percentages of RET in several kind of anaemia were statistics significant(P0.01).Conclusion The percentage of CD55-/CD59-in patients with IDA,AA,AA-PNH increased distinctly,especially in the patients with PNH obvious.It would be helpful for the diagnosis,antidiastole and prognosis when combination of CD55/CD59and RET.
