Abstract
Objective To evaluate the significance of deficiency of CD55/CD59 on peripheral blood cells in differentiation and diagnosis of PNH and anemia.Methods CD55/CD59monoclonal antibodies labeled by fluorescence were used to analyze percentage of CD55/CD59 on erythrocyte and neutrophil in 30 normal subjects,32 cases of AA,28 cases of PNH or AA-PNH,38 cases of IDA,24 cases of MA and 12 cases of AIHA patients with flow cytometry.Results The percentage of CD55/CD59 on erythrocyte and neutrophil in normal subjects were less than 5%,in PNH or AA-PNHS patients were more than 20% and in some of AA patients were more than 5%(They were all less than 15%).The percentage of CD55-/CD59-on erythrocyte in about 40% IDA patients were more than 5%(They were all less than 20%),while on neutrophil were less than 5%.The percentage of CD55-/CD59-on erythrocyte and neutrophil in MA and AIHA patients were all less than 5%.Conclusion The peripheral blood cells CD55/CD59 deficiency on erythrocyte and neutrophil analyzed by FCM is the most reliable and sensitive test in differentiation and diagnosis of PNH,AA-PNH,AA,IDA,MA and AIHA.【
