STUDIES ON ELISA METHOD FOR DETECTING Vibrio alginolyticus, THE PATHOGENIC BACTERIA OF Pseudosciana crocea

Abstract

Vibrio alginolyticus, the pathogenic bacteria of Pseudosciana crocea was inactivated by the addition of formaldehyde to 0. 05% (30 ℃, 8 h) for vaccine. Anti- V. alginolyticus serum was obtained from rabbits by injecting vaccine. The anti serum has no cross reaction to V.parahaemduticus and other nine strains after being absorbed. Anti serum wes used for V. alginolyticus detecting by ELISA method. The lowest V. alginolyticus suspension detectable was 97 OOO cfu/ ml. This ELISA method was used for pathogenic bacteria detecting in aquatic farming P.,crocea. The results showed that V. alginolyticus in seawater was undetectable; 80% liver of diseased P. crocea were positive in V. alginolyticus while in the normal, 20%. The results indicated that ELISA method could be used not only to diagnose the clinically diseased P. crocea, but also to recognize the carrier.
