Abstract
Objective To explore the protective effect of DSLZ on acute chemical liver injury induced by 1% carbon Tetrachloride(CCl4)in mice and to reveal its mechanism.Methods 60 mice were randomly divided into normal control group,model group,SalviolⅡ A group,high dose DSLZ group(2.6mg/kg),middle dose group(1.56mg/kg)and low dose group(0.52mg/kg).All the mice were intragastrically administered the corresponding agents respectively for 30 days.After the last administration 1% CCl42.0ml/kg was given to mice in all groups except control group in a single administration of vegetable oil intragastrically to establish the acute liver injury mouse model.16 hlater,the mice were sacrificed,alanine aminotransferase(ALT),aspartate aminotrsferase(AST),malondialdehyde(MDA),reduced glutathione hormone(GSH)level in serum were detected.HE staining was performed for observing the pathological changes of the liver.Results Compared with the control group,the level of ALT,AST,MDA in the serum of the model group mice were increased significantly,and the GSH level was decreased significantly(P0.01).Compared with model group,the serum ALT,AST,MDA level in hign dose of DSLZ and middle dose of DSLZ groups were decreased significantly(P0.05 or P0.01),while the GSH level in serum were increased significantly(P0.05 or P0.01).Moreover the HE staining results showed that in model group the swelling,kytoplasm coacervation,hydropic degeneration and necrosis of liver cells were seen.The morphological damage of liver cells were improved significantly in DSLZ groups.Conclusion DSLZ has a significant protective effect on CCl4–induced acute liver injury,which might be related to the anti-oxidation.
